亚低温降低创伤后脑组织炎症反应的实验研究

摘要 【目的】研究亚低温在外伤致脑组织炎症反应中的治疗作用。【方法】液压冲击法建立实验动物脑创伤模型,分创伤组和亚低温治疗组两组,亚低温治疗组动物接受冰袋全身降温处理;RT-PCR及Western blotting法检测环氧化-酶2(COX-2)mRNA及蛋白水平表达,ELISA法检测前列腺素E2(PGE2)水平。【结果】创伤组脑组织COX-2 mRNA、蛋白表达水平于伤后早期明显升高,第3天达高峰,此后逐渐降低,其反应产物PGE2出现相同变化趋势。亚低温治疗组脑组织COX-2 mRNA、蛋白表达,及其产物PGE2水平与创伤组伤后同时期比较降低明显,差异具有统计学意义(P<0.05)。【结论】亚低温可明显降低外伤后脑组织炎症因子的产生和释放,降低炎症反应程度。 [Objective] To study the treatment effect of hypothermia on inflammatory reaction after brain injury. [Methods] Animal models of traumatic brain injury were established in rat, and the rats were divided into two groups, including traumatic group and hypothermia-treated group. After hypothermia-treated, brain tissues were taken and mRNA and protein level expression of cydooxygeaase-2 (COX-2), were detect with RT-PCR and Western blotting to and prostaglandin-E2 (PGE2) level was detected by enzyme linked immunosorbent assay (ELISA). [Results] The results from RT-PCR and Western Blot indicated that the expression of COX-2 in mRNA and protein level in traumatic group were increased obviously after brain injury, and the PGE2 level increased too. However the mRNA and protein level of COX-2 in hypothermia-treated group were decreaced significantly than control group at the same time point, as well as the PGE2 changed. [ Conclusions] The hypothermia treatment conld suppressive the express of inflammatory factors obviously, and depress the extent of inflammatory reaction after brain injury.
