用mRNA差异显示技术(mRNAdifferentialdisplay)对分别用200U/ml重组人IFN-a和IFN-Υ处理的MA891肿瘤细胞系总mRNA进行分析,分离出10个差异表达的片段,并进行了克隆。因IFN-a抑制MA891细胞系的肺转移能力,而IFN-Υ则增强其肺转移能力,故认为该10个片段所代表的基因中可能有某个或几个基因与肿瘤转移有关。
We have previously reported that in vitro treatment with interferons of MA891, a spon-taneous mouse mammary tumor capable of metastasizing to the lungs upon subcutaneous in-oculation, could modulate its metastatic potential- IFN-Y increases while IFN-a decreaseslung metastasis. Identifying the genes differentially expressed in MA891 cells separatelytreated with IFN-a and IFN-Y. MA891 cells were treated in vitro with 2ooU/ml IFN-a orIFN-Y respectively for 48h. Total RNA was isolated with the single step method of acidguanidinium thiocyanate. After being digested by DNase I, the two RNA samples were dif-ferentially displayed with Tl2MA/T/C/G X 20 arbitrary primer pairs. Most of the RT-PCRwere done twice. The differentially expressed cDNA bands were isolated, reampliated andcloned. Ten differentially expressed cDNA bands were identified: T2a,T2Y,C4a,T6a,T1lY,G15Y,T16Y, T5a1, T5a2,T17a, T15a, T15Y, named according to the type of IFN and primerpairs used. The total RNA samples of MA891 cells treated by IFN-a and IFN-Y were differ-entially displayed with T12MA/T/C/G X 20 arbitrary primer pairs. Theoretically they cancover 95 % mRNAs- It is anticipated that one or more of the ten cDNA clones may be relatedto the metastatic potential of MA891 cells.
