脑通有效部位组方对大鼠脑缺血—再灌注损伤保护作用的机制研究

摘要 目的:研究脑通有效部位组方对大鼠脑缺血—再灌注损伤保护作用的机制。方法:采用大鼠大脑中动脉阻塞造模,分为3组:即假手术组,模型组,脑通给药组,观察脑通主要有效部位组合对脑缺血再灌注损伤大鼠脑组织NO含量、NOS活力、DNA电泳和Bcl-2表达的影响。结果:脑通组NO含量、NOS活力显著降低,DNA电泳条带变暗,Bcl-2表达升高。结论:脑通主要有效部位组合可能是通过降低脑组织中NO含量,抑制NOS活性,增强Bcl-2蛋白的表达,抑制细胞凋亡,起到保护脑缺血再灌注损伤的作用。 Objective: To study the protective mechanism of active fractions of naotong on injury of cerebral ischemia reperfusion in rats. Methods: Models of cerebral ischemia were made by blocking middle cerebral arteries; then divided the rats of cerebral ischemia randomly into the sham-operated group, model group, and naotong treated group. Concentration of NO, activity of NOS, DNA agarose gel electrophoresis and expression of Bcl- 2 of the rats cerebra were observed for the effect and protective mechanism of active fractions of naotong on injury of cerebral ischemia reperfusion. Results: In the naotong treated group, concentration of NO and activity of NOS decreased, the band of DNA electrophoresis darkened, the expression of Bcl-2 increased. Conclusion: The protective mechanism of active fractions of naotong on injury of cerebral ischemia reperfusion in rats may lie on decreasing NO, inhibiting the activity of NOS and the apoptosis of cells, increasing the expression of Bcl-2 in cerebrum.
