摘要
建立了高效液相色谱法检测植物性原料中三聚氰胺的方法。样品由水涡旋混匀超声提取,通过加入亚铁氰化钾和乙酸锌沉淀蛋白进行净化。色谱柱SunfireC_18(4.6×250mm,5μm),流动相为水-氨水-高氯酸缓冲液,检测波长240nm,检测限0.5mg/kg。在添加浓度为1.0~4.0mg/kg范围内,回收率在70.2%~80.2%,相对标准偏差小于10%。
An analytical method for determination for melamine in vegetable materials by high performance liquid chromatog- raphy was developed. Samples were extracted by blending with water. The extract was purified by adding potassium hexaoyan oferrate (Ⅱ) trihydrate and zinic acetate dihydrate to precipitate protein. The HPLC conditions: PR18(4.6 × 250 mm, 5 μm), water-ammonia-perchloric acid as mobile phase, and UV detection at 240 nm. The limit of detection was 0.5 mg/kg. The average recoveries were 70.2 %-80.2 % at the concentration of 1.0-4.0 mg/kg. Relative standard deviation was less than 10%.
