摘要
对本室已构建的汉滩病毒糖蛋白(GP)和核蛋白(NP)的嵌合基因G1S0.7重组腺病毒的免疫学特性进行研究。将汉滩病毒含有G1S0.7嵌合基因的重组腺病毒直接免疫Balb/c小鼠,用ELISA、微量细胞培养中和实验及淋巴细胞增殖实验检测小鼠的免疫应答效果。用该重组腺病毒免疫的小鼠,可诱导产生抗汉滩病毒NP及GP特异性的抗体,抗体效价分别为1∶320及1∶40。同时重组腺病毒还可刺激机体产生低水平的中和抗体和明确的淋巴细胞增殖反应。说明所构建的汉滩病毒嵌合基因G150.7重组腺病毒,可同时刺激机体产生特异性的抗汉滩病毒体液免疫应答及细胞免疫应答。
To investigate the immunological properties of recombinant adenovirus containing hantaan virus chimeric gene G1 SO. 7. Balb/c mice was directly immunized by recombinant adenovirus G1 SO. 7. The immune response was tested by ELISA, microcell-culture neutralizing experiment and T lymphocyte proliferation test (MTr assay). The results showed that the chimeric gene G1SO. 7 can stimulate combining antibodies against NP (1:320) and GP (1:40) respectively detected by ELISA, which suggested that both GP and NP immune response were activated. The low-titer neutralization antibody reaction and clear lymphocyte proliferation were also detected simuleity. It is suggested that the recombinant adenovirus G1 SO. 7 can directly stimulate Balb/c mice to produce specific humoral immunity and cellular immunity to against hantaan virus.
