汉防己甲素防护环孢素A肾毒性的实验研究 被引量:12

Experimental study on the protective effect of tetrandrine on cyclosprine A nephrotoxity in rats

摘要 本文观察了中药钙拮抗剂汉防己甲素对大鼠环孢素A肾毒性(CsA-NT)的防护作用。雄性Wistar大鼠用CsA50mg·kg-1/d灌胃7天导致急性CsA-NT,表现为血清肌酐(Cr)升高34.8%,内生肌酐清除率(Ccr)下降39.4%,组织学检查发现近曲小管大片空泡变性。同时,肾皮质丙二醛(MDA)含量显著升高,超氧化物歧化酶(SOD)活性显著下降。大鼠每日用CsA灌胃前30分钟,给予汉防己甲素10mg/kg腹腔注射,能使上述指标明显改善,组织学损伤也明显减轻。实验结果提示汉防己甲素能从功能和形态上防护大鼠急性CsA-NT。 The present experimental study investigates the effect of tetrandrine, a calcium antagonist extracted from the Chinese medicinal herb, on cyclosporine A nephrotoxity (CsA-NT) in the rats. Administration of CsA 50 mg.kg-1/d. to a male Wistar rat by gavage for 7 days resulted in acute CsA-NT, with a 34. 8% increase in serum creatinine and a 39. 4% decrease in creatinine clearance (Ccr). Histological examination demonstrated an extensive vacuolization in proximal tubule. In addition, renal cortical malondialdehyde (MDA) was increased significantly, while the activities of renal cortical superoxide dismutase (SOD) decreased noticeably. In protective experiment, tetrandrine (10mg. kg-1 /d, i. p) was administered approximately 30 min prior to CsA.Seven days later, in coadministered tetrandrine with CsA groups, the values of the parameters mentioned above, as well as histological damage, were significantly improved. It was indicated that tetrandrine could functionally and structurally prevent acute CsA-NT in rats.
