反相高效液相色谱法同时测定黄芪中的4种酚酸 被引量:6

Simultaneous Determination of Four Phenoic Acids in Astragalus by Reversed-Phase High Performance Liquid Chromatography

摘要 建立了黄芪中4种酚酸类化合物的高效液相色谱测定方法,并分析比较了提取方法和测定条件,实验色谱条件为:ZORBAX Eclipse XDB-C8柱(150mm×4.6mm;5μm),以乙腈-0.4%冰醋酸水溶液(体积比为22:78,pH=2.9)为流动相,柱温:28℃,流速:0.8mL/min,检测波长为325nm和260nm,外标法定量,样品加标回收率在89.34%-103.45%,相对标准偏差小于4.5%,该法简便、快速、灵敏,可用于黄芪中4种酚酸的同时测定。 Four phenoic acids in Astragalus were determined by reversed-phase high performance liquid chromatography, and the methods of the extraction and determination of the samples were analyzed and compared. The following optimal method was identified. The samples were analysed by a ZORBAX Eclipse XDB-C8 column (150mm×4.6mm;5μm). The mobile phase was acetonitrile-0.4% acetic acids (22 : 78, v/v, pH=2.9). Column temperature was 28℃. The flow rate was 0.8 mL/min. DAD-detection was per-formed at 325 nm and 260 nm. Spike recoveries of samples ranged from 89.34%-103.45% with a rela- tive standard deviation of 〈4.5 %. The proposed method is simple and sensitive and can he used to simul- taneously determine four phenoic acids in Astragalus.
