摘要
目的观察中药双蒲散对慢性萎缩性胃炎模型大鼠的治疗作用,并探讨其作用机制。方法大鼠以饮用0.1%氨水建立慢性萎缩性胃炎(CAG)模型,治疗90d后观察胃黏膜炎症和组织病理学情况,检测大鼠胃黏膜血流量(GMBF)、胃黏膜超氧化物歧化酶(SOD)活力、谷胱甘肽(GSH)、丙二醛(MDA)及前列腺素(E2PGE2)含量,胃液酸度和胃蛋白酶活性。结果双蒲散大、中剂量组大鼠胃黏膜组织学接近正常组。与模型组相比,双蒲散各剂量组大鼠胃黏膜SOD、GSH、PGE2明显升高,MDA显著降低,GMBF显著增高,大鼠胃液酸度和胃蛋白酶活性明显提高(P<0.05,P<0.01)。结论双蒲散对慢性萎缩性胃炎模型大鼠胃黏膜有良好的治疗作用,能改善和逆转实验性萎缩性胃炎大鼠胃黏膜萎缩。其机制可能与其增加胃黏膜血流量、增强胃黏膜屏障功能、调节胃酸和胃蛋白酶分泌、抗自由基损伤等有关。
Objective To study function of Shuang Pu Powder on Treating Chronic Atrophic Gastritis （CAG） model rats and investigate Its mechanism. Methods Make rats CAG model with 0.1%ammonia water, after 90 days treated ,observe the gastric mucosa inflammation and gastric atrophy level,detect the change of glutathione（GSH）, superoxide dismutase （SOD）, prostaglandinE2（PGE2）, malonyl dialdehyde （MDA） contents in gastric mucosa.measuring Gastric mucosal blood flow （GMBF） , secretion of gastric acid and pepsin activity, spleen weights and weight indexes , thymus weights and weight indexes. Results There is no marked meaning to the differentiation of gastric mucosa pathematology and microanatomy among the Shuang Pu Powder high-dosage ,medium-dosage groups and the normal control group. Compared to model group, Shuang Pu Powder has maked a significant increase of SOD , GSH and PGE2 in the CAG rats, and can make the levels of MDA decrease significantly, and can make a significant increase of GMBF,Shuang Pu Powder has maked a significant increase of secretion of gastric acid and pepsin activity,and can increased the weight, spleen weights indexes and thymus weight indexes of rats （P〈 0.05,P〈 0.01）. Conclusion The Chinese drug Shuang Pu Powder has good curative effect to chronic atrophic gastritic model rats,and it can also improve and reverse the gastric atrophy of experimental atrophic gastritis of rats.Its mechanism maybe related with improving blood circulation of gastric mucosa, strengthening gastric mucosal protective screen, regulating gastric acid and pepsin activity, resisting the freeradical impairment and so on.
