摘要
目的研究槲皮素联合TNP-470对肺腺癌小鼠移植瘤组纵中内皮抑素(Endostatin,ES)和血管内皮细胞生长因子(vascular endothelial growth factor,VEGF)表达的影响。方法复制肺腺癌LA795细胞的T739小鼠异体移植瘤模型。接种后4 d随机分为4组,每组10只。对照组:生理盐水0.2 ml/只,于接种后第4 d开始腹腔注射,1次/d,共16次;TNP-470组;TNP- 470(30 mg·kg^(-1)·d^(-1))溶于0.2 ml丙二醇中,于接种后第4 d开始腹腔注射,隔日1次,共7次:槲皮素组:槲皮素(5 mg·kg^(-1)·d^(-1))溶于0.2 ml生理盐水中,余同对照组;联合组:按TNP-470组和槲皮素组用药。肿瘤接种后24 d,处死后剥离皮下肿瘤称湿重后固定标本,用免疫组化和图像分析系统定量检测肿瘤组织ES和VEGF的表达,剖胸观察肺转移结节数。结果各治疗组肿瘤湿重和肺转移结节数均低于对照组,其中以联合组最低(P<0.01);联合组移植瘤组织中ES表达最高(P<0.01),而VEGF表达最低(P<0.01)。肺转移结节数和瘤湿重均与ES呈负相关,与VEGF则呈正相关。结论槲皮素协同TNP-470可能通过促进ES表达,抑制VEGF表达,从而抑制肿瘤生长转移。
Objective To study the effects and significance of quereetin and TNP-470 on ES and VEGF of the transplanted tissues in lung adenocarcinoma mice. Methods LA795 cells were subcutaneously inoculated into the mammary fatty pad of T739 mice to clone lung adenocarcinoma model. Four days after inoculation, 40 mice were randomized into four groups (10 mice in each group): control group in which every mouse was peritioneally injected with 0.2 ml 0.9% sodium chloride aqua every day, 16 times in all; TNP-470 group in which every mouse was injected with TNP-470 for (30mg·kg^-1·d^-1) next days, 7 times in all; quercetin group in which every mouse was injected with quercetin (5 mg·kg^-1·d^-1) once a day, 16 times in all; Alliace group in which: the dose is consisted with those in quercetin group and TNP-470 group. After transplantation and all mice were sacrificed after 24 days, lung metastatic occurrence rat were counted and wet weight after peeling the subcutaneous tumor was measured, fix the specimen of tumor, immunohistochemistry method with image analysis system was applied to detect the ES and VEGF in tumor tissues. Results The lung metastatic and wet weight in every treatment group was significantly lower than those in the control group, the lowest in the Alliace groups, with a significance; the effect of ES in tumor tissue of very treatment group was higher than that of the control groups; the highest in the Alliace group, with a great difference; the expression of VEFG in tumor tissue of very treatment group was lower than that of the control groups; the lowest in the Alliace groups, with a significant difference; the effect of ES has a negative relationship with the effect of VEFG, the lung metastatic and wet weight, the effect of VEFG had a negative relationship with the lung metastatic and wet weight. Conclusion Quercetin and TNP-470 may inhibit expression of VEGF, promote expression of ES, hence inhibits growth and metastasis of tumor.
