摘要
目的探讨小剂量氯化镉对颌下腺颗粒曲管(GCT)细胞线粒体的损伤及丙酸睾酮的保护作用。方法35只雄性Wistar大鼠分为3组:对照(C)组、镉(Cd)组和镉加丙酸睾酮(CA+T)组。利用透射电镜作颌下腺GCT细胞线粒体的超微结构观察,并利用image-plus-pro全自动图像分析系统作线粒体损伤的定量研究。结果镉注射后24h,GCT细胞内出现不同程度的线粒体肿胀和(或)空泡化,胞质和线粒体内可见髓样结构,7~15d后上述改变明显加重,30d后细胞形态仍未恢复正常。Cd+T组的超微结构损伤较相应CA组轻。CA组在镉注射后3~15d,Gcr细胞线粒体的各项指标与对照组相比均有显著差异(P〈0.05)。CA+T组仅在镉注射后7d和15d,GCT细胞线粒体比表面显著低于对照组(P〈.05)。结论镉对GCT细胞线粒体的超微结构有损伤作用,补充雄激素对上述损伤有保护作用。
Objective To Investigate the impaired effects of low-dose cadmium chloride and the protective effects of testosterone propionate on mitochondria of CJCT cells in submandibular gland of male rats. Methods Thirty-five male adult Wistar rats were randomly divided into three groups: control group, Cd injecting group and Cd + T injecting group. The morphologic changes of GCT cells were observeded by transmission electron microscopy, and the quantitative analysis were performed on mitochondria with image-plus-pro analysis system. Results The mainly ultrastructural changes after injecting Cd in 24 hours included swelling and/or vacuolation of mitochondria, increasing of myelin figures in cytoplasm and mitochondria of GCT cells, which were most prominent in 7-15d, moreover, the changed morphology of GCT cells didn't recover in 30d. On the other hand, the ultrastructural lesions of GCT cells in Cd+T group were gentler compared with Cd group. In image analysis, there was significant difference between Cd group and control group in 3- 15d (P;0.05), and between Cd+T group and control group in 7d and 15d (P;0. 05). Conclusions The present study indicate that the ultrastructure of mitochondria will be injured by cadmium. Furthermore, supplying testosterone can prevent the mitochondria in GCT cells from toxicity of cadmium.
