摘要
本研究观察了二氧化氮(NO2)对大鼠肺脂质过氧化、抗氧化酶及花生四烯酸代谢的影响。实验选用Wistar雄性健康成年大鼠,体重150~175g,设计浓度:自然空气,0.15,1.5,15mg/m3NO2组。暴露方法:静式染毒,每天2小时,每周6天,染毒4周。实验结果表明:(1)随着NO2浓度的递增,大鼠尿中血栓烷素B2与6-酮-前列腺素F1α比值逐渐升高,且存在剂量-反应关系。(2)1.5mg/m3NO2能致大鼠肺丙二醛含量增加,同时可见谷胱甘肽过氧化物酶活性升高,且两者相关具有高度显著性(P<0.01)。这表明:大鼠肺能通过某些抗氧化酶活性的升高而免受脂质过氧化物的毒害。(3)1.5mg/m3NO2能致大鼠肺血栓烷素B2增加,6-酮-前列腺素F1α降低。同时可见丙二醛与6-酮-前列腺素F1α之间存在显著负相关(P<0.05)。经对尿中血栓烷素B2,6-酮-前列腺素F1α及其比值与肺中相应指标作相关分析,各指标相关具有显著性。表明:测定尿中血栓烷素B2与6-酮-前列腺素F1α含量的变化能反映NO2对大鼠肺花生四烯酸代谢的影响,用它作为一个能反映NO2健康效应、灵敏的生化指标成为可能。
The effects of NO 2 on rats were studied in vivo,by measurement of molnodialdehyde(MDA),glutathione peroxidase(GSH PX),superoxide dismutase(SOD),6 keto prostaglandine F 1(6 keto PGF 1),thromane B 2(TXB 2).We choiced specitic pathogen free male wistar rats,weighting 150 ̄175g,exposed to either room air or 0.15,1.5,15mg/m 3 NO 2 in an 250L exposure chamber for 4 weeks,2 hours/per day,6 days/per week.Our results showed that 1.5,15mg/m 3 NO 2 could elevated significantly ratios of urinary TXA 2/Keto PGF 1α .It indicated that there was dose effect relationship between NO 2 and urinary AA metabolites.In the experiment of effects of 1.5mg/m 3 NO 2 on rat′s lungs,our results showed that NO 2 could significantly increased the amounts of MDA and GSH PX activities in lung homogenates on 4th,7th day respectively it showed a significant positive correlationship between MDA and GSP PX( P <0.01).It is possible that the rats could defend lung cells against cytotoxic lipid peroxide by increaseing some antioxidative protective enzymes,such as GSH PX and also found that the contents of TXB 2 increased on 7th and the contents of 6 keto PGF 1 deseased significantly on 4th,7th,14th.We found that it showed a significant negative correlationship between MDA and 6 keto PGF 1α ( P < 0.05 ).Comparing the pulmonary TXB 2,6 keto PGF 1α with urine′s,there is significant positive correlationship( P < 0.05). This showed that changes of urinary TXB 2,6 keto PGF 1α may reflect the effects of NO 2 on AA metabolism of lung well,and be as a good,early and sensitive biochemical indicator for healthy effects.
