冠心病患者循环中EPCs与VEGF的关系及临床意义 被引量:9

Relationship between vascular endothelial growth factor and circulating endothelial progenitor cells in patients with coronary artery disease and its clinical significance

摘要 目的:研究冠心病患者循环中血管内皮生长因子(VEGF)与内皮祖细胞(EPCs)的关系及临床意义.方法:55例入选者均经冠状动脉造影根据有无冠状动脉狭窄而分为两组:冠心病组30例,对照组25例.采集入选者外周血,体外培养扩增内皮祖细胞,7d后通过倒置相差显微镜计数内皮祖细胞克隆形成单位(EPC-CFU)评估外周血EPC水平,同时用ELISA法检测血浆中VEGF水平,分析冠脉组病人VEGF与EPCs的关系.结果:冠心病组VEGF水平低于对照组[(19.6±6.9)pg/mLvs(35.4±9.4)pg/mL,P<0.001)];冠心病组血循环中EPC-CFU数量也较对照组降低(12.4±4.2vs19.0±5.1,P<0.001);血循环EPCs数量与VEGF浓度成正相关(r=0.493,P<0.01).结论:VEGF的下降可能是导致血循环中EPCS的减少的原因,可能参与了冠心病的发生和发展. AIM: To investigate the correlation between vascular endothelial growth factor (VEGF) and circulating endothelial pro- genitor cells (EPCs) in coronary artery disease (CAD) patients and its clinical significance. METHODS: Fifty-five patients were divided into two groups : 30 patients with coronary artery dis- eases (CAD) and 25 control patients with angiographically normal coronary arteries. After 7 days of culture,the number of EPCs col- ony-forming units (EPC-CFU) was counted under phase-contrast microscope. The plasma level of vascular endothelial growth factor (VEGF) was determined by ELISA. Relation between the level of VEGF and the number of EPCs was analyzed in patients with CAD. RESULTS : The level of VEGF in the CAD group was significantly lower than that in the controls [ ( 19.0 ± 3.8 ) pg/mL vs (34.2±4.6) pg/mL,P 〈0. 001 ]. The number of EPC-CFU in CAD group was significantly lower than that in the controls (9.8±2.9 vs 18.9 ±4.3 P〈0.001). A positive correlation between the plasma level of VEGF and the number of EPC-CFU(r = 0. 493, P 〈 0.01 ) was also observed. CONCLUSION: The decline of VEGF may induce the decrease of circulating EPC and be involved in the occurrence and development of coronary heart diseases.
