摘要
目的:制备抗创伤弧菌mAb并用于创伤弧菌感染的快速诊断.方法:用灭活创伤弧菌免疫Balb/c小鼠,采用杂交瘤技术制备抗创伤弧菌的mAb,ELISA法用于mAb的Ig效价,亚类及特异性鉴定.结果:获得了2株可持续分泌抗创伤弧菌mAb的杂交瘤细胞株.结论:获得的高特异性的抗创伤弧菌mAb不仅为创伤弧菌感染的快速诊断奠定了坚实的基础,而且为其进一步的治疗方面的研究提供了理论依据.
AIM: To prepare monoclonal antibody ( mAb ) against Vibrio vulnificus and evaluate its roles in rapid diagnosis for Vibrio vulnificus infection. METHODS: Balb/c mice were immunized by inactived Vibrio vulnificus. Anti-Vibrio vulnificus McAbs were prepared by using hybridoma technique. The ELISA was used to identify Ig subgroup, titers of ascites, relative affinity and the specificity of McAbs. RESULTS: Two cell lines of hybridoma were established and they constantly secreted high quality McAbs steadily. CONCLUSION: The McAbs established could be highly specific for Vibrio vulnificus, which not only lays a solid foundation for development of a rapid method for detection of Vib- rio vulnificus infection but also provides the theoretical basis for further investigation of the treatment.
