摘要
从无锡惠山采集的土样中分离获得一株过氧化氢酶产量较高的菌株SYBC-01,根据其形态、生理生化特性和16S rDNA序列分析,鉴定该菌为粘质沙雷氏菌(Serratia marcescens)。初步试验表明,该菌产过氧化氢酶酶活达293.88 U/mg细胞,所产过氧化氢酶的最适反应pH 7,最适反应温度70℃,在pH6-10范围内较稳定,60℃以下热温度性较好,表观Km值为23.71 mmol·L^-1,Vmax为34 672 U·mg^-1蛋白。
A strain with high capability of producing catalase was isolated from in the soil of Huishan, Wuxi. According to the morphological, physiological characterizations, and its 16S rDNA sequence, the strain was identified as Serratia rnarcescens. The data of preliminary experiment showed that the yield of catalase production from Serratia SYBC-01 reached 293.88 U/mg, the optimal pH and temperature was 7 and 70℃ respectively. The enzyme was stable under pH6-10 and below 60℃. The apparent Km and Vmax value was 23.71 mmol·L^-1 and 34 672 U·mg^-1 respectively.
