常态虚寒状态小鼠CYP3A活性P-gp水平比较研究 被引量:7

A Comparative Study on the Level of P-glycoprotein and the Activity of Cytochrome P4503A of Normal and Deficient Cold Condition of Mouses

摘要 目的:比较虚寒状态与常态小鼠肝和小肠细胞色素P4503A(CYP3A)活性和P-糖蛋白(P-gp)水平情况。方法:肌注氢化可的松复制小鼠虚寒状态模型,采用聚乙二醇法制备肝和小肠微粒体,测定肝和小肠细胞色素CYP 3A活性;以免疫组化法检测肝和小肠组织P-gp水平。结果:虚寒状态组和正常状态组肝和小肠微粒体内CYP3A活性和P-gp水平存在差异,即正常状态CYP3A活性、P-gp水平高于虚寒状态,且差异显著(P<0.05)。结论:初步发现,CYP3A活性和P-gp水平存在机体状态差异。 Objective: To observe the level of P -glycoprotein and the activity of eytoehrome P4503A in liver and small intestine between Normal and Deficient Cold Condition of mouses. Methods : The model of Deficient Cold Condition was replicated with Hydrocortisone intramuscular injection . The liver and small intestine mierosomes were prepared with polyethylene glycol and the activities of CYP450 3A in the mierosomes were determined by the substrate of erythromyein and the level of P - glycoprotein in liver and small intestine tissue by SP staining methods of Immunohistoebemistry. Results ; The level of P - glycoprotein and the activity of cytoehrome P4503A in liver and small intestine of Normal condition group is significantly higher than that of Deficient Cold Condition group. Conclusion: The level of P- glyeoprotein and activity of eytoehrome P4503A are different in different orgarlic condition.
