人ω干扰素克隆、表达及体外抑制冠状病毒昆徐株作用的研究

摘要 目的制备人ω干扰素(hIFN—ω)并初步研究其体外抗普通冠状病毒活性。方法制备hIFN—ωcDNA,并在大肠杆菌BL-21中高效表达、纯化后,测定了hIFN—ω的抗滤泡口膜炎病毒(VSV)和冠状病毒昆徐株活性。结果成功地获得了hIFN—ωGST融合蛋白,利用WISH—VSV系统检测hIFN—ω的抗病毒活力为7.81×10^4kU/L;与基因工程α干扰素(IFN—α2b)的比较发现,hIFN—ω可以明显抑制冠状病毒昆徐株的活性,而IFN—α2b未见此活性。结论hIFN-ω具有抑制冠状病毒昆徐株的活性,有独特的抗呼吸道病毒作用;不同的干扰素亚型对不同的病毒敏感性有所差异。 Objective To obtain the human interferon-ω and observe its anti-ωronavirus activities. Methods The cDNA of human interferion-ω was obtained,and the IFN-ω fusion protein was expressed and purified in Escherichla colt(E, coli) BL-21. The anti-virus activities of human interferon-ω were accessed by using WISH-VSV system,and then compared with those of IFN-α2b by using coronavirus(Kunxu strain). Results GST-IFN-ω fusion protein was obtained successfully. And the anti-virus activity of IFN-ω was 7.81× 10^4 kU/L, which was accessed by WlSH-VSV system. Compared with IFN-α2b, IFN-ω inhibited cytopathic effect of coronavirus (Kunxu strain) , and IFN-α2b had no such effect. Conclusions IFN-ω could inhibit coronavirus, indicating that in may have special way on anti-respirovirus. Also sensibilities were different among interferon subtypes.
