摘要
[目的]研究大肠癌组织中β-catenin、cyclinD1和c-myc蛋白的表达及临床意义。[方法]采用免疫组化法检测正常大肠黏膜和大肠癌组织中β-catenin、cyclinD1和c-myc蛋白的表达。[结果]正常大肠黏膜β-catenin蛋白表达定位于细胞膜,大肠癌组织β-catenin细胞浆和细胞核表达(异位表达)明显增加,细胞膜表达有不同程度的减少。大肠癌组织异位表达率为63.1%,细胞膜表达缺失率为70.8%,大肠癌组织β-catenin的异位表达率与肿瘤分化程度、临床分期和淋巴结转移有关。大肠癌组织中cyclinD1和c-myc表达明显高于正常大肠黏膜。大肠癌组织中β-catenin异位表达率与cyclinD1和c-myc阳性表达率均呈明显正相关。[结论]异常的β-catenin相关信号通路与大肠癌的发生发展有密切的关系,β-catenin的异常表达可能可以作为大肠癌患者诊断及预后评估的良好指标。
[Purpose] To investigate the expression and clinical significance of β-catenin, cyclinD1 and c-myc in eolorectal cancer(CRC). [Methods] The expression of β-catenin, cyclinD1 and c-myc in normal colorectal mucosa and CRC was detected by immunohistochemistry method. [Results] In normal colorectal mucosa, β-catenin was localized on the cell membrane. Increased expression on cytoplasmic and nuclear and reduced expression on membrane were detected in CRC. The cytoplasmic and nuclear expression rates of β-catenin was 63.1% in CRC. The negative membrane expression rate of β-catenin was 70.8% in CRC. The cytoplasmic and nuclear expression rate of β-catenin was associated with histological differentiation, clinical stage and lymph node metastasis in colorectal carcinoma. The positive expression rates of cyclin D1 and c- myc in the CRC were significantly higher than those in normal colorectal mucosa. The cytoplasmic and nuclear β-catenin expression rate was significantly correlated with the positive expression rate of cyclin D1 and c-myc in the CRC. [Conclusions] The abnormal β-catenin correlated signaling pathway plays a critical role in the progression of CRC. The abnormal expression of β- catenin might be a useful index for diagnosing and predicting prognosis for CRC.
