卡介苗配用腺苷对小鼠的免疫接种效果

摘要 目的探讨腺苷(ADO)提高卡介苗(BCG)免疫接种的效果。方法实验第2天,取小鼠接种部位皮肤与其脾细胞共育3d,测定其上清液细胞因子浓度和腺苷脱氨酶(ADA)活性。实验第4周,小鼠经气道给予减毒结核分支杆菌(MTB)1×10^6 CFU,18—20h后检测其血清ADA活性、肺免疫细胞类型、巨噬细胞(AM)溶酶体活性变化及脾细胞活化数。结果①局部接种部位皮肤与脾细胞共同培养3d,ADO组TNF—α水平显著高于对照组,ADO组ADA活性明显低于对照组,而两组间IFN-γ、IL-13无明显差异;②MTB感染肺部后,ADO组AM数量及溶酶体活性均明显高于对照组,ADO组血清ADA活性显著低于对照组,而中性粒细胞、淋巴细胞、及脾活化细胞数两组均无明显差异。结论卡介苗配用ADO能促进小鼠免疫接种效果。 Objective To investigate the efficacy of adenosine promoting the bacillus ealmette guerin vaccine (BCG) immune inoculation. Methods On the second day of the test, the skin inoculated with BCG vaccine was taken and co cultivated three days with spleen ceils. The activity of ADA and the concentration of cytokines in the culture supernatant were detected respectively. On the fourth week, attenuated myeobaeterium tuberculosis was given with the dose of 1 ×10^6 CFU through airway for both groups . Then we detected the activity of serum ADA, the type of lung immune cell, the activity change of maerophage lysosome and the activation status of spleen cells. Resuits The concentration of TNF-α in ADO group was significantly higher than that in control group and the activity of ADA was statistically lower than that in control group. While no significant differences of IFN-γ/and IL-13 coneentration were found in two groups. (2) The number of maerophage and the activity of lysosome of the lung in ADO group were significantly higher than those in the control group respectively after the lung was stimulated by MTB, whereas the activity of Serum ADA was significantly lower in ADO group than that in control group. There were no significant difference of the number of neutrophils, lymphoeytes and spleen cells in two groups. Conclusion Adjuvant ADO can enhance anti-bacterial effect of macrophage against mycobacterium tuberculosis.
