摘要
目的:探讨急性胰腺炎(AP)肺组织中巨噬细胞移动抑制因子(MIF)的表达及其与核因子-κB(NF-κB)的关系。方法:30只SD大鼠随机分为正常对照组、急性水肿型胰腺炎(AEP)组和急性坏死性胰腺炎(ANP)组。以不同浓度牛磺胆酸钠液逆行胰胆管插管注射造成大鼠AEP和ANP两种模型。6 h后检测相关指标。应用免疫组化染色检测肺组织NF-κB的表达;采用逆转录-聚合酶链反应(RT-PCR)技术,对肺组织MIF的表达水平进行半定量测定;生化法检测髓过氧化物酶(MPO)活性;行肺、胰腺组织病理学检查并评分。结果:正常对照大鼠肺组织中MIF mRNA和NF-κB呈低表达。AP模型组大鼠肺组织中MIF mRNA的表达显著增强,NF-κB、MPO活性和病理学评分也显著增高,且在ANP组增高尤为明显;两个模型组间各项指标具有显著性差异(P<0.05)。MIF mR-NA的表达与NF-κB的活性密切相关。组织学评分显示,随着胰腺损伤的加重,肺损伤也加重。结论:MIF参与了AP肺损伤的发病过程,其机制可能与激活NF-κB有关。
Objective: To explore the expression of macrophage migration inhibitory factor and its relationship with nuclear factor-κB(NF-κB) in acute pancreatitis-associated lung injury. Methods: Thirty SD rats were randomized into the normal group, acute edematous pancreatitis (AEP) group, and acute necrotizing pancreatitis (ANP) group. AEP and ANP were induced by the retrograde injection of sodium taurocholate into pancreatobiliary ducts. All the animals were sacrificed at 6 h after induction of pancreatitis. Activation of NF-κB in pulmonary tissues was determined with immunohistochemistry. The expression of MIF in pulmonary tissues was detected by semi-quantitative reverse transcription polymerase chain reactive (RT-PCR). Myeloperoxidase (MPO) activity was detected by biochemistry and the histopathologic scores of pancreases and lungs was evaluated. Results: The expression of MIF mRNA and NF-κB was low in normal group. And the expression of MIF mRNA, the activity of NF-κB and MPO and the histopathologic scoring of pancreases and lungs in rats of AP model groups increased significantly (all P〈 0.05), especially in ANP group. All the parameters of the two AP model groups were statistically different(P〈0.05). The expression of MIF mRNA was closely related to NF-κB activation. The histologic scores demonstrated an increasing damage of lung. Conclusion: MIF could be in volved in the pathogenesis of sodium taurocholate-induced acute pancreatitis-associated lung inju ry and may correlate with NF-κB activation.
