摘要
目的:探讨雌激素对人正常皮肤成纤维细胞的影响,为进一步探讨雌激素抗皮肤衰老的机制和开发抗衰老护肤品提供新思路。方法:采集正常成人皮肤标本,分离培养真皮成纤维细胞;以流式细胞术检测17β-E2作用前后成纤维细胞内活性氧(ROS)水平的改变,以及检测丙二醛(MDA)含量和三种抗氧化酶(SOD,GSH-px,Cat)的活性。结果:①雌激素浓度为1×10-6-1×10-10mol/L(生理浓度至药理浓度范围之内)时,可促进人皮肤成纤维细胞的增殖;②雌激素浓度为1×10-7-1×10-10mol/L时,可以减少人正常皮肤成纤维细胞内MDA的生成,加速三种抗氧化酶的产生;③1×10-6-1×10-10mol/L雌激素作用于人皮肤成纤维细胞后,可以降低细胞内活性氧(ROS)的产生。结论:雌激素在一定浓度时具有改善人正常皮肤成纤维细胞生物学特性的作用,且药物浓度与对人皮肤成纤维细胞的作用密切相关。
Objective: To study the effects of estrogen on the biological characteristics of human skin fibroblasts in vitro. Methods: Human dermal fibroblasts were isolated by trypsin digestion method from normal adult skin specimens and cultured in DMEM containing 10% newborn bovine serum. The anti-oxidative activity was evaluated with biochemical methods and the level of reactive oxygen species (ROS) was detected with flow cytometry. Results: (1) A significant increase of proliferation was observed in skin fibroblasts after cultured with estrogen at the dosage of 1 × 10^-6-1 × 10^-10mol/L; (2) Estrogen could reduce the MDA expression and accelerate the expression of cellular superoxide dismutase, glutathione peroxidase and Catalaseare in skin fibroblasts;(3) Estrogen could obviously reduce the level of ROS in skin fibroblasts in the dosages of 1 × 10^-6-1 × 10^-10 mol/L. Conclusion: Estrogen could improve the biological characteristics of human skin fibroblasts in vitro. Our investigation provides cellular laboratorial evidences for the medical cosmetology to apply estrogen in order to postpone skin aging.
