摘要
目的探讨小鼠视网膜缺血再灌注(RIR)损伤后铜蓝蛋白在视网膜中的表达和意义。方法采用前房灌注法建立RIR损伤动物模型,免疫组织化学方法结合计算机图像分析系统定性和半定量分析不同时段视网膜组织中铜蓝蛋白的表达。结果正常小鼠视网膜仅在神经节细胞层可见铜蓝蛋白表达,缺血再灌注损伤后6h,铜蓝蛋白开始在内丛状层、内核层和外丛状层表达并逐渐增加,48h达高峰,之后下降。结论铜蓝蛋白参与了RIR损伤的早期过程,其作用可能是清除自由基和保护视网膜。
Objective It is reported that oxygen-derived free radicals play an important role in retina ischemia-reperfusion injury. Ceruloplasmin is thought to be an important enzymatic antioxidants. This study tried to investigate the expression of ceruloplasmin in ischemia-reperfusion retina of mouse. Methods The model of experimental retinal ischemia-reperfusion injury was induced by irrigation of balance saline solution in the anterior chamber of the right eyes to increase the intraocular pressure to 120 mmHg for 60 minutes in 24 BALBIC mice. The expression of ceruloplasmin in retina of model mouse was quantitatively analyzed 1,6,12,24,48 and 72 hours by immunohistochemistry. The content of ceruloplasmin in retina of mouse was semi-quantitatively analyzed at various time points of modeling using computer image analysis system. Results Immunohistochemistry analysis revealed that ceruloplasmin was expressed in retinal ganglion cell layer other than other retinal layers in normal BALBIC mouse. The ceruloplasmin was expressed at 6 hours after modeling and the positive cells located in retinal inner plexiform layer, inner nuclear layer and out plexiform layer of retina. The computer image analysis showed that expression of ceruloplasmin was increased with time lapse of retinal ischemia-reperfusion injury with the peak expression at 48 hours of modeling and decreased gradually 72 hours later. Conclusion Ceruloplasmin participates in retinal ischemiareperfusion injury by eliminating free radical in retina and protecting the retina from the ischemia-reperfusion injury consequently.
