大分子右旋糖酐致血瘀大鼠体表特征研究 被引量:2

Surface characteristics in rats with blood-stasis syndrome induced by macromolecule dextran

摘要 目的探讨大分子右旋糖酐致血瘀大鼠体表特征的变化规律。方法将64只大鼠随机分为8组,即模型组的30、60、120、240 min组与对照组的30、60、120、240 min组,每组8只。模型组大鼠尾静脉注射用0.5%的伊文思蓝溶液配制的10%的大分子右旋糖酐溶液,对照组大鼠尾静脉注射用生理盐水配制的0.5%的伊文思蓝溶液,造模后30、60、120、240 min时观察并拍摄各组大鼠耳廓及舌的颜色变化,测量其血浆及耳廓提取液中伊文思蓝含量,进行统计学分析。结果模型组大鼠造模30 min后耳根及舌尖出现蓝色瘀斑,随着时间的延长,蓝色瘀斑范围扩大,240 min时全耳廓呈现深蓝色,舌体瘀斑范围>1/2,对照组耳廓始终半透明,舌体颜色均匀;模型组血浆伊文思蓝含量明显低于对照组;模型组耳廓伊文思蓝含量明显高于对照组。结论静注大分子右旋糖酐所致血瘀证大鼠耳和舌的体表特征明显异于对照组,基本符合中医关于血瘀证的描述。 Objective To investigate the change law of surface characteristics in rats with blood-stasis syndrome induced by macromolecule dextran. Methods All rats (n = 64) were randomly divided into 8 groups, including the model groups ( groups of 30 min. , 60 min. , 120 min. and 240 rain. , each n = 8) and control groups (groups of 30 rain. , 60 min. , 120 min. and 240 min. , each n =8). The rats in the model groups were injected 10 % macromolecule dextran solution confected by 0. 5 % Evens blue solution through the caudal vein, and the rats in the control groups were injected 0.5% Evens blue solution confected by normal saline solution. The color changes of rat auricle and tongue in different groups were observed and photoed 30, 60, 120 and 240 minutes after the model establishment. The contents of Evens blue in rat serum and auricle extract were detected and analyzed statistically. Results In the model groups the blue petechia in rat auricle and tongue were observed after 30 minutes, which aggravated as the time went by. After 240 minutes the whole ear of rat changed to dark blue and the blue petechiae of tongue enlarged to more than half of tongue. In the control groups the rat auricle was semitransparent and the tongue color was normal. The contents of serum Evens blue in the model groups were much lower than those in the control groups. The contents of auricle Evens blue in the model groups were much higher than those in the control groups. Conclusion Surface characteristics of ear and tongue in rats with blood-stasis syndrome induced by macromolecule dextran is different significantly to those in the rats of control group, which accords with the description of blood-stasis syndrome in TCM.
