摘要
目的研究血管内皮细胞生长因子反义寡核苷酸(VEGF-ASODN)对裸鼠荷人胃癌肿瘤的生长抑制作用.方法人工合成硫代磷酸化VEGF-ASODN,建立人胃癌荷瘤裸鼠移植瘤模型(共3组),实验分为4组,(1)VEGF-ASODN组:于瘤体及瘤周注射VEGF-ASODN66μg;(2)错义寡核苷酸组:注射错义寡核苷酸66μg;(3)生理盐水组:注射生理盐水,每3d1次,共治疗7次;(4)另设无荷瘤对照组.实验结束后2d,处死动物,取血清测VEGF蛋白,测量肿瘤大小及重量,取肿瘤组织用流式细胞技术测增殖细胞核抗原(PCNA)及HE切片染色检查.结果VEGF-ASODN能抑制VEGF的表达,显著降低荷瘤裸鼠血清VEGF蛋白水平,减小肿瘤的体积和重量,PCNA表达量VEGF反义治疗组76.16%,错义寡核苷酸组87.50%,生理盐水组91.16%.结论VEGF-ASODN通过抑制VEGF表达,从而能抑制胃癌细胞的生长、增值.
Objective To observe the inhibitory effect of VEGF antisense phosphorothioate oligodeoxynucleotides on the growth of human gastric cancer xenografts in nude mice. Methods The VEGF-ASODN was synthesized artificially. After the model of human gastric cancer xenografts in nude mice were founded, they were randomly divided into three groups: antisense group, scrambled group and saline group and control group was also established. Antisense (66 μg), scrambled sequence (66 μg) and saline (once every three days and 7 times in all) were injected in the first three groups, respectively. Two days after the therapy, the mice were sacrificed, serums were used for detection of VEGF protein, the tumor were measured and weighted, PCNA was detected by FCM. Results The VEGF-ASODN could suppress the expression of VEGF in human gastric cancer xenografts in nude mice and reduce VEGF protein in serum of nude mice significantly. It could also reduce the volume and weight of xenografts. The expression of PCNA was 76.16% ,87.50% and 91.16% in antisense group, scrambled group and saline group, respectively. Conclusion VEGF, VEGF-ASODN can inhibit proliferation of human gastric cancer xenografts By inhabiting the expression of in nude mice.
