实用光老化动物模型建立方法的探讨 被引量:32

摘要 目的:探讨光老化动物模型造模过程中的各个关健因素,确定实用的造模方法。方法:用电推剪剃除大鼠背部预定部位的毛,将大鼠置于专用固定器中,每天10min,用SS-03AB型紫外线光疗仪(UVA:340~400nm;UVB:260~350nm)连续照射60天,第61天处死大鼠,照射部位皮肤组织切片进行HE染色,观察造模效果,并结合实践总结造模经验。结果:与空白组大鼠皮肤相比较,模型组呈现表皮层厚薄不一,真皮层血管扩张,大量炎性细胞浸润,胶原纤维明显变性等典型光老化特征,造模成功。结论:本实验中讨论并确定的造模关健因素简便易行,经济可靠,可复制性强,为光老化动物模型的复制建立了一种实用的方法。 Objective To ascertain a practical method of constructing rats photoaging model, the key factors were studied. Methods The dorsal hair of rats was shaved by electrical clipper. The dorsal hairless rats were fixed and exposed to ultraviolet A (UVA340- 400nm)and ultraviolet B (UVB260- 350nm) for 60 days (10min a day) successively. In the 61st day, all the rats were killed and histological evaluation (HE strain) of irradiated skin was carried out. Results Compared to control group, the epidermis thickness of irradiated group was not uniform, dermis vascellums were dilated, inflammatory cells infiltrated and collagen fibers were metamorphotic. Conclusion The rats photoaging model was copied successfully and the key factors were investigated. This method is easy, economic and repeatable. It can be generally used for photoaging study.
