SVZa神经干细胞诱导分化为多巴胺能神经元的实验研究 被引量:4

摘要 目的研究骨形成蛋白-2(bone morphogenetic protein-2,BMP2)及星形胶质细胞条件培养液(astrocyte-conditioned medium,ACM)在室管膜前下区(anterior subventricular zone,SVZa)神经干细胞分化为多巴胺能神经元中的作用。方法体外培养SVZa神经干细胞,然后分为对照组、BMP2组、ACM组、BMP2+ACM组,通过免疫组化和流式细胞仪技术检测各组中SV-Za神经干细胞分化为TH+(酪氨酸羟化酶)细胞的比例。结果各实验组的TH+细胞比例均明显高于对照组,其中BMP2+ACM组分化比例最高,差异有统计学意义(P<0.05)。结论BMP2及ACM可以促进SVZa神经干细胞向多巴胺能神经元分化,而且BMP2与ACM有正协同作用。 Objective To study the role of bone morphogenetic protein-2(BMP2) and astrocyte-conditioned medium (ACM) in SVZa neural stem cells differentiating into dopaminergic neurons. Methods The neural stem cells of SVZa were separated and cultured in vitro, which were divided into: control group, BMP2 group, ACM group, BMP2 + ACM group. The TH + cells rate of differentiation of the neural stem cells of SVZa was detected by immunohistochemistry and flow cytometry (FCM). Results The rate of TH+ cells differentiation in each experiment group were much higher than control group, especially in the group of BMP2 + ACM. There was significant difference among all groups by analysis of variance(P〈0. 05). Conclusion BMP2 and ACM can promote the dopaminergic neuronal differentiation of the neural stem cells of SVZa and have positive cooperativity.
