摘要
研究了核糖胞嘧啶和脲嘧啶共聚物的酶促合成条件。共聚物PolyCxU与PolyI在类似生理盐水条件下形成错配的双链RNA,然后对PolyI:C和错配的双链RNA对核酸酶的敏感性及紫外熔化曲线进行了比较研究。
The enzymic synthesis conditions of Cytosine Uracil copolymers have been investigated. The equivalent molar weight mixtures of Poly CxU and Poly I in physiological saline like conditions formed the mismatched double stranded RNA. The comparable studies of Poly I:C and Poly I:CxU on the sensitive to nucleases and UV melting curves have shown that the sensitivity to nucleases for Poly I:CxU(in case of x>7) increased by 2-5 fold and the melting points decreased to 4-10℃ .A hypothesis that I and U bases occupied in the double stranded RNA could be in status wobble base pairs(I·U) has been suggested.
