甲醛和苯对小鼠睾丸总抗氧化能力和ATP酶及钙的影响 被引量:6

摘要 目的探讨甲醛和苯对小鼠睾丸总抗氧化能力和ATP酶及钙的影响及二者联合毒效应的类型。方法采用昆明种雄性小鼠54只随机等分为9组,按3×3析因设计要求设9个染毒组,甲醛和苯染毒剂量分别为0、5、10mg/kg和0、100、400mg/kg,连续灌胃染毒5d,35d后处死小鼠取睾丸制作匀浆液测Na+-K+-ATP、Ca2+-Mg2+-ATP酶活性、Ca2+含量和总抗氧化能力。结果甲醛和苯各染毒组小鼠睾丸总抗氧化能力、Na+-K+-ATP、Ca2+-Mg2+-ATP酶活性和Ca2+含量与对照组比较差异有显著性(P<0.01),并有量-效关系,甲醛和苯之间的联合毒性效应是协同作用。结论甲醛和苯能抑制小鼠睾丸Na+-K+-ATP、Ca2+-Mg2+-ATP酶的活性、降低总抗氧化能力和提高Ca2+的含量,二者对这些指标的影响表现为协同毒效应,这可能是甲醛和苯对生殖功能损伤的机制。 Objective To study the combined toxicity effects of formaldehyde and benzene on the total antioxidant capacity, ATPase, and Ca^2+ in the testicle of mice. Methods Fifty- four Kunming male mice were randomly divided into 9 treated groups. Various doses of formaldehyde (0,5, and 10 mg/kg), benzene (0, 100, and 400 mg/kg) separately or formaldehyde plus benzene were given into 9 groups of mice by gastric gavage for .5 successive days. After 3.5 days, these mice were sacrificed to measure the activity of Na^+ - K^+ - ATPase, the activity of Ca^2+ - Mg^2+ - ATPase, the content of Ca^2+ and the total antioxidant capacity in testicle of mice. 3 × 3 factorial analysis was used in the experiment. Results Compared with the negative control group, the activity of Ca^2 - Mg^2+ - ATPase, the activity of Na^+ - K^+ - ATPase, the total antioxidant capacity and the content of Ca^2 + of the mice testicles among treatment groups presented a significant dose - effect relationship and significance of difference (P〈 0.01 ). And the joint toxic effect exhibited synergistic effect. Conclusions In the testicle of mice, the activity of Na^+ - K^+ - ATPase, the activity of Ca^2+ - Mg^2+ - ATPase and the total antioxidant ca- pacity can be restrained, while the content of Ca^26 can be increased by formaldehyde and benzene, which shows the testicular cells of mice can be enhanced to damage jointly by formaldehyde and benzene.
