摘要 目的观察孕酮对脑缺血后脑水肿的影响,并从孕酮对血-脑屏障通透性的影响和抗氧自由基两方面进一步探讨其可能的作用机制。方法雄性SD大鼠72只随机分为假手术组、I/R组、孕酮组。进行脑组织含水量测定,伊文思蓝渗透法血-脑屏障通透性定量分析,电镜下血-脑屏障超微结构形态学观察,脑组织SOD活性和MDA含量测定。结果孕酮能明显减轻脑水肿,降低血-脑屏障通透性,抑制SOD活性的降低和MDA含量的增高。结论孕酮抑制氧自由基的损害,降低血-脑屏障通透性,减轻脑缺血后脑水肿。 Objective To observe the effect of progesterone on cerebral edema during cerebral ischemia/reperfusion injure, and to approach its mechanism from both BBB ( blood brain barrier) and free radical. Methods 72 male rats were randomly divided into three groups: sham -operation groups, model groups and progesterone groups. To observe the water content of the brain, and evaluate the functional changes with content of permeated Evens Blue, ultramicrostructure of BBB through electron microscope and assaying of SOD and MDA in the brain. Results Progesterone can obviously decrease the permeability of BBB, alleviate the degree of edema, increase the activity of SOD and lower the contents of MDA. Conclusions Progesterone can inhibit the damage of oxygen free radical, decrease the permeability of BBB, and alleviate the degree of edema after cerebral ischemia.
