摘要 目的:探讨来氟米特(leflunomide,LEF)在体外对人外周血T淋巴细胞的免疫调节作用。方法:在植物血凝素(phytohemagglutinin,PHA)刺激下,人外周血淋巴细胞与不同剂量的LEF(5,15,45μg/ml)共培养。检测各组T淋巴细胞的增殖、凋亡情况,分析各组T淋巴细胞免疫表型CD152,CD28的表达以及各组T淋巴细胞的IFN-γ,IL-10,TGF-β1 mRNA水平。结果:在体外,LEF对由PHA诱导的T淋巴细胞的增殖、凋亡均有抑制作用,且抑制作用与LEF呈剂量依赖性。LEF对T淋巴细胞CD152,CD28的表达无明显影响。LEF能促进IL-10,TGF-β1 mRNA的表达,抑制IFN-γmRNA的表达。结论:LEF可能通过抑制T淋巴细胞增殖、减少T淋巴细胞凋亡、促进T淋巴细胞IL-10、TGF-β1 mRNA表达及抑制IFN-γmRNA表达来下调免疫反应。 Objective: To investigate the immunoregulatory effects of leflunomide ( ripheral of T-lymphocytes in vitro. Methods: The LEF (5,15,45 μg/ml) were added lymphocytes from unrelated donors in the presence of phytohemagglutinin(PHA). We LEF) on human peinto wells containing assessed the proliferation of T-lymphocytes. The immunophenotypes ( CD152, CD28 ) of T-lymphocytes were analyzed by flow cytometry. We also observed the T lymphocytes' apoptosis and the mRNA expression of interferon-γ (IFN-γ), interleukin-10 (IL-10), transforming growth factor-β1 (TGF-β1). Results: In all the experiments, with the presence of LEF human peripheral T-lymphocytes resulted in a statistically significant decrease of their proliferative activities and apoptosis as dose-dependent manner. No inhibitory effects on CD152 and CD28 of T-lymphocytes had been observed. LEF promote the mRNA expression of IL-10, TGF-β1 and inhibit IFN-γ. Conclusion: LEF can inhibit the proliferative activities and apoptosis of human peripheral T cells and promote the mRNA expression of T-lymphocytes' IL-10, TGF-β1 and can lower the mRNA expression of IFN-γ, which may play an important role in its immunoimpressive affects.
