摘要
目的建立兔高胆固醇-动脉粥样硬化模型,观察阿托伐他汀对脑组织血管紧张素Ⅱ(AngⅡ)和血红素氧合酶-1(HO-1)表达的影响。方法24只健康新西兰白兔随机分为3组:正常对照组,高脂饮食组,高脂饮食加阿托伐他汀组。喂饲8周后,取血测定血清总胆固醇(TC)及低密度脂蛋白胆固醇(LDL-C)水平,取脑组织检测AngⅡ和HO-1的表达。结果(1)AngⅡ的表达:高脂组明显高于对照组,他汀组表达较高脂组明显降低(P<0.05);(2)HO-1的表达:高脂组较对照组明显升高,他汀组较高脂组进一步升高(P<0.05)。结论高胆固醇血症时脑组织肾素血管紧张素醛固酮系统(RAAS)处于激活状态,阿托伐他汀通过抑制AngⅡ的生成并相应增加HO-1的表达,对机体起到抗动脉粥样硬化的保护作用。
Objective To establish model of rabbit with hypercholesterolemia and investigate the influence of Atorvastatin on the expression of angiotensinⅡ (AngⅡ) and hemo oxygenase-1 (HO-1) in rabbit brain tissue. Methods Twenty-four healthy New-Zealand albino rabbits were divided into 3 groups randomly., normal control, high cholesterol diet, high cholesterol diet and Atorvastatin. After feeding for 8 weeks, fasting blood samples were collected and total cholesterol(TC)and low density lipoprotein cholesterol (LDL-C) levels were determined. Rabbit brains were extracted after sacrifice and expressions of AngⅡ and HO-1 were assayed. Results AngⅡ expression in high cholesterol diet group was higher than in control group (P〈0.05) ; Ang Ⅱ expression in Atorvastatin group was lower than in high cholesterol diet group (P〈0.05). HO-1 expression in high cholesterol diet group was higher than in control group (P〈0.05) ; HO-1 expression in Atorvastatin group was higher than in high cholesterol diet group (P〈0.05). Conclusions Rennin-angiotensin aldosterone system (RAAS) in brain tissue is activated under high cholesterol diet. Atorvastatin can prevent atherosclerosis by reducing Ang Ⅱ production and increasing HO-1 expression.
