摘要
目的应用微波处理ＡＢＣ免疫组化法，对福尔马林固定、石蜡包埋切片组织内的ＤＨ７抗原性恢复进行研究。方法微波热时间分别以７、１４、２１、２８ｍｉｎ处理组织切片。ＤＨ７单抗分别以１２０、１４０、１８０、１６０四种稀释度测试。结果微波热１４ｍｉｎ，ＤＨ７单抗稀释度１８０，染色结果良好。结论实验结果提示：既往认为长期福尔马林液固定石蜡包埋的组织不宜免疫组化染色，而通过微波处理恢复其组织抗原性，仍可获得良好的染色结果。此种恢复组织抗原性的方法简便，可减少免疫组化的假阴性率，并可适用于外检材料有关免疫组织化学的回顾性研究。
Objective To test the effect of microwave on retrieval of DH7 antigen in formalin fixed and paraffin embeded tissue by applying avidin biotin complex (ABC) immunohistochemical technique. Methods Microwave heatingtime of 14 min and a optimal dilution of 1:80 DH7 monoclonal antibody were found to be the choice in a pilot study. Avidin biotin complex tech nique was used to detect DH7 antigen in gastric carcinoma specimens. Results Good DH7 immunostaining results were obtained after heating by microwave for 14 min. and with a dilution of 1:80 monoclonal antibody of DH7. Conclusion These results indicated that following the retrieval of antigen DH7, adequate staining could be achieved for tissues being formalin fixed and paraffin embeded for long time which were previously considered unsuitable for immunohistochemical analyses. This simple and practical procedure for antigen retrieval could obviously for antigen retrieval could reduce the incidences of false negative immuno staining results, and, furthermore,it might be applied for retrospectivestudy of those stored formalin fixed and paraffin embeded specimens.
