摘要
用还原性烟酰胺腺嘌呤二核苷酸磷酸黄递酶,简称黄递酶(reducednicotinamidedinu-cleotidephosphatediaphorase,NADPH-d)组织化学的方法,在光镜下对NADPH-d反应阳性细胞进行计数和计算机图像分析测定反应产物的光密度(opticaldensity,OD)值,观察一氧化氮合酶(NOS)阳性神经元及纤维在惊厥鼠上丘分布的情况。实验动物为听源性惊厥易感性大鼠,简称惊厥鼠(P77PMC)和正常Wistar大鼠,简称正常鼠。实验结果如下:(1)在大鼠上丘第Ⅰ层NOS阳性神经元呈密集均匀分布,在第Ⅳ层其分布也是均匀的但较稀疏,第Ⅶ层只有少量NOS阳性神经元散在分布于中部。第Ⅰ、Ⅳ、Ⅶ层NOS阳性神经元的大小和形态是一致的。在第Ⅰ、Ⅶ层NOS阳性神经元呈上丘的背腹方向排列。在第Ⅳ、Ⅵ层有密集的NOS阳性纤维。(2)上丘NOS阳性神经元的数目以在急性发作的惊厥鼠最多,其次为非急性发作的惊厥鼠和电铃刺激的正常鼠,而在无电铃刺激的正常鼠最少。(3)NOS阳性反应的OD值在各组大鼠间无显著性差异。本研究结果显示,上丘NOS阳性神经成分的分布有特异性;NOS酶的活性在惊厥鼠是增高?
The reduced nicotinamide dinucleotide phosphate diaphorase(NADPH d)histochemistry method was performed to study the laminar distribution of the nitric oxide synthase(NOS)activity and the morphology of positive neurons in the superior colliculus of the P77PMC rats and the normal Wistar rats.The optic density(OD) of the reactive products were measured with the computor linked image analysis system(IBSA2000,OPTON).The following results were obtained:1.In the layⅠⅠ and Ⅳ the positive neurons were evenly distributed,but densely in layer ⅠⅠ and loosely in layer Ⅳ.In layer Ⅶ only a few NOS positive neurons were sparsely located in the central portion.In the lay ⅠⅠ and Ⅶ,the positive neurons were arranged in the dorsoventral direction of the superior colliculus.The positive neurons in different layers were morphologically unaminous.In the layer Ⅳ and Ⅵ there were densely NOS positive fibers distributed.2.The number of the positive neurons in layer ⅠⅠ,layer Ⅳ and Ⅶ was more in the P77PMC rats which were submitted to the acoustic stimuli than in the other groups.The positive neurons were fewer in the non auditory stimulative normal Wistar rats than in the other groups.3.The OD value of the reactive products in the layerⅠⅠ was not significantly different among the four groups,neither were in the other layers of the superior colliculi.Our results showed that the NOS containing neurons were distributed in the particular regions of the superior colliculus,and the NOS positive neurons were significantly more in the P77PMC rats than in the normal Wistar rats.
