摘要
目的:探讨肾虚型骨性关节炎造模方法的可行性和有效性。方法:将12只4月龄未孕SD雌性大鼠随机分为模型组、对照组,模型组行双侧卵巢切除去势结合单侧膝前后交叉韧带、内侧副韧带切断,对照组仅行单侧膝前后交叉韧带、内侧副韧带切断。比较术后第2、4、6周膝关节软骨大体形态和病理学评分、雌二醇含量变化。结果:模型组软骨组织病理学评分显著高于对照组(P<0.05)。结论:采取双侧卵巢切除去势结合单侧膝前后交叉韧带、内侧副韧带切断的方法,建立动物模型能有效反映肾虚型骨性关节炎的退变情况,适合作为肾虚型骨性关节炎的动物模型进一步运用推广。
Objective: To investigate the feasibility and effectiveness of the method for establishing rat models of kidney - deficiency osteoarthritis (OA). Methods : Twelve 4 - month - aged non - pregnant SD rats were equally randomized into model group and control group. The model group received bilateral ovariectomy, and the cutting - off of unilateral medial collateral ligament, anterior cruciate ligament, and posterior cruciate ligament. The control group received the cutting - off of unilateral medial collateral ligament, anterior cruciate ligament, posterior cruciate ligament only. After operation for 2, 4 and 6 weeks, the scores of macroscopic and pathological features of knee joint cartilage as well as the eslradiol content were observed. Re-sults: The score of pathological score of knee joint cartilage in the model group was higher than that in the control group ( P < 0. 05). Conclusion: The modeling method of bilateral ovariectomy, and the cutting-off of unilateral medial collateral ligament, anterior cruciate ligament, and posterior cruciate ligament can establish the rat models of kidney - deficiency osteoarthritis successfully, and is worth of further extension.
