摘要
利用以乙醇为夹带剂的CO2超临界萃取技术(SFE-CO2)萃取黄蜀葵花有效成分并进行工艺优化;研究萃取物镇痛活性.以液固比、压力、乙醇浓度、CO2流量、温度5个影响因素进行正交实验,优化工艺.用小鼠扭体法来证明其镇痛药理活性.最佳工艺条件为:液固比3.0 mL/g,萃取压力30 MPa,乙醇浓度90%,CO2流量4 kg/h,萃取温度65℃,乙醇与粉末直接混合加入、萃取时间1.5 h,浸膏得率为5.43%,黄酮得率为0.29%,萃取物的镇痛率达60.7%.用SFE-CO2(乙醇)萃取法得到的黄蜀葵花萃取物具有较好镇痛效果,该法是一种较优越的黄蜀葵花萃取方法.
The effective components of Abelmoschl manihot L medic were extracted by the technique of supercritical CO2 extraction with ethanol as a cosolvent. The extracting conditions were optimized. And the analgesic activity of the extracted product was studied. Five factors which affect extraction ( extraction temperature, pressure, the concentration of ethanol cosolvent, the flux of CO2, the volume of ethanol) were investigated and optimized by an orthogonal design. The writhing test in mice was used to examine the analgesic activity. The optimum extracting conditions were: pressure 30 MPa, temperature 65 ℃, time 1. 5 h, concentration of ethanol 90%, the volume of ethanol 3.0 mL/g, and the flux of CO2 4 kg/h. Under the above conditions, the extraction rate was 5.43%. The analgesic effect of the extracted product (0. 8 mg/kg, ip) was 60. 7% tested by writhing test in mice. The supercritical CO2 extraction is a good process for Abelmoschl manihot L medic extraction.
