不同pH下10种鱼尿酸酶活性的比较研究 被引量:2

摘要 为寻找一种新的理化性质与人体内环境比较接近的尿酸酶,对10种鱼肝组织尿酸酶活性进行了测定,并对其最佳提取pH进行了比较。结果显示:10种鱼肝组织在pH6.8~11.1范围内均可测到尿酸酶活性,其酶活性范围为0.064~0.644U;在pH6.8、7.5、8.4和pH11.1条件下鲢(Hypophthalmichthys molitrix)、翘嘴红鲌(Erythroculter ilishaeformis)、鳜(Siniperca chuatsi)、团头鲂(Megalobrama am-blycephala)、黄颡鱼(Pseudobagrus fulvidraco)、鲶(Silurus asotus)、银鲫(Carssius auratus gibelio)、鲤(Cyprinus carpio)、鳙(Aristichthys nobilis)、草鱼(Ctenopharyngodon idellus)肝组织中尿酸酶平均酶活力分别为0.405、0.356、0.306、0.287、0.278、0.204、0.168、0.128、0.123、0.094U。在pH7.5缓冲液抽提条件下,活性较高的是翘嘴红鲌(0.402U)、团头鲂(0.301U)、黄颡鱼(0.297U)。以肉食性鱼类或以动物性食物为主食的鱼类肝组织尿酸酶活性较高,以植物性食物为主食的鱼类肝组织尿酸酶活性较低。翘嘴红鲌、团头鲂和黄颡鱼尿酸酶的理化性质比较适合人体内环境;与猪比较(猪肝组织平均酶活力0.206U),鱼肝组织中的尿酸酶催化尿酸氧化反应的酸碱稳定pH范围较大,特别是在pH7.5条件下仍具有明显活性。该结果可为今后筛选临床应用的尿酸酶参考。 In order to search for a kind of uricase,whose property is close to the inner condition of human body,the uricase activity in livers of ten species of fish were determined and the optimum pH of extracting were compared.The results showed that the uricse activity could be detected in all of the livers of ten fish in the pH range from 6.8 to 11.1,and the uricase activity scope was 0.064~0.644 U (wet weight).Under four kinds of pH condition,the average uricase activity of Hypophthalmichthys molitrix,Erythroculter ilishaeformis,Siniperca chuatsi,Megalobrama amblycephala,Pseudobagrus fulvidraco,Silurus asotus,Carssius auratus gibelio,cyprinus carpio,Aristichthys nobilis,Ctenopharyngodon idellus were 0.405,0.356,0.306,0.287,0.278,0.204,0.168,0.128,0.123 U and 0.094 U respectively.Under the condition of pH 7.5,the uricase activity of Erythroculter ilishaeformis,Megalobrama amblycephala and Pseudobagrus fulvidraco are relatively higher than others,being 0.402,0.301,0.297 U respectively.The results of above indicated that the uricase activity in the livers of flash-eater fish was relatively higher,while the activity of urecase was lower in plant eating fishes.The pH range,in which fishes uricase can be soluble,were wider than that of pigs (average activity of uricase is 0.206 U),especially in the pH 7.5.The results would be useful in the selection of clinical appliable uricase in the future.
