高压氧对体外培养成骨细胞分化的影响 被引量:6

摘要 目的探讨高压氧治疗对成骨细胞分化的影响。方法将体外培养的成骨细胞接种于96孔培养皿中,每孔10000个细胞,正常培养3d后,改用成骨化培养基,24h后,成骨细胞接受2.4个大气压90min和1.5个大气压90min的高压氧和高压空气的治疗,每天1次,共19次。结果高压氧治疗组第3d可以检测出钙沉积的出现,而对照组直到第7d才能检测出钙沉积的出现。钙沉积分析表明,从第3d开始到第19d,高压氧治疗组的钙沉积都显著高于对照组。在成骨化培养条件下,高压空气治疗后,钙沉积没有明显增加。在高压氧治疗后的第5、9、13d进行的碱性磷酸酶的活性分析显示,高压氧治疗组的碱性磷酸酶活性也显著高于对照组。Von-Kossa染色和显微镜下观察显示,高压氧治疗增加了骨结节的形成。结论高压氧治疗能够明显刺激成骨细胞的成骨分化。 Objective To investigate the effect of hyperbaric oxygen (HBO) on the differentiation of osteoblasts in vitro. Methods Osteoblasts were seeded in 96 well plates at a cell density of 10 000 cells per well. After 3 days normal culture, medium was changed to osteogenic medium. Subsequently, cultures were exposed daily to HBO of 2.4 ATA for 90 minutes and 1.5 ATA for 90 minutes up to 19 days. Mineralization was evaluated by calcium deposition assay, alkaline phosphatase (ALP) activity and yon Kossa staining. Results Significant calcium deposition was observed after 3 days of HBO treatment and the calcium deposited was consistently higher in HBO treatments period, there was no difference in the amount of calcium deposition between the treatment of hyperbaric air and untreated control group. In addition, ALP activity and the biomarker for osteogenic differentiation were also significantly increased within the HBO-treated cultures, yon Kossa staining showed that bone nodule formation of HBO group was more than the control group. Conclusions HBO treatment could significantly stimulate osteogenic differentiation, which implies a potential application of HBO in bone tissue engineering.
