摘要
目的:观察肾组织超微结构变化及部分凋亡基因表达,探讨中药驱铅丸对慢性铅中毒大鼠肾保护作用的部分机理。方法:在饮水中添加0.02%醋酸铅,连续60天制作大鼠慢性铅中毒模型,然后将模型动物随机分为4组:中药高剂量治疗组(中药驱铅丸3.5g/kg/天灌胃)、中药低剂量治疗组(驱铅丸0.7g/kg/天灌胃)、Ca-EDTA治疗组(依地酸钠钙加普鲁卡因肌肉内注射,50mg/kg/天,连续注射4天为1个疗程,休息4天后进行下一个疗程,连续治疗7个疗程)及模型组(不予任何治疗),连续60天。观察治疗前后血、尿及肾组织中铅(Pb)等元素含量变化,并检测肾组织P53、bcl-2mRNA表达及超微结构改变。结果:模型动物体重、血红蛋白降低;经过治疗后,各组尿Pb、血Pb含量显著降低(P<0.05),尿钙尿锌降低,肾钙肾锌增高(P<0.05);各治疗组bcl-2表达增高(P<0.05,P<0.01),P53表达降低(P<0.01),中药高剂量治疗组P53mRNA表达低于低剂量组和Ca-EDTA治疗组(P<0.05),bcl-2mRNA表达高于中药低剂量组和Ca-EDTA治疗组(P<0.05);电镜结果显示治疗组肾小管上皮细胞胞浆内质网增多,偶见致密小体,基底膜侧微粒增加,细胞器间隙稍肿胀,与模型组比较更趋于正常。结论:中药驱铅丸能降低血液及肾组织Pb含量,促进Pb排出;通过调节P53介导bcl-2产生的细胞凋亡过程而发挥对铅中毒造成的肾组织损害的保护作用。
Objective:To explore the pathological change of kidney's ultrastrue and the expression of some apoptosis genes of traditional Chinese medicine QQP(QuQian Pill)to kidney's protection in rat with chronic lead toxicosis.Method:Build up the chronic lead toxicosis model with drink the water with 0.02% lead acetate 60 days,then the model rats were randomly divided into 4 groups:the low QQP dosage group(8 rats)added the QQP 0.7g/kg/d from intragastric administration,the high group was 3.5g/kg/d,the Ca-EDTA group(edetate calcium disodium)was 50mg/kg/d from intramuscular injection,and model group nothing.After 60 days,the change of part element in the blood,urine,bone and kidney between before and after treatment,the expression of P53,bcl-2 mRNA and kidney's ultrastructure were measured.Results:Compared with not drink contained-lead water,weight and hemoglobin were down;after treatment,lead level in urine,blood and kidney significantly decreased(P<0.05),calcium and zinc in urine fall down,calcium and zinc in kidney were up(P<0.05).After treatment,expression of bcl-2 increased,but expression of P53 decreased(P<0.05).The expression of P53 mRNA in TCM high-dose group was lower than in low-dose group and Ca-EDTA treatment group;the expression of bcl-2 mRNA was higher than in low-dose group and EDTA treatment group.Electron microscope showed that,compared with the model group,endoplasmic reticulum in renal tubule epithelium cytoplasm increased,with little pyknotic bodies,granules beside the basal membrane increased,organelle spatium became swollen,it was subject to the normal.Conclusion:Qu-lead bolus can reduce the lead level in the blood and kidney,promote elimination of lead;it protect the kidney through regulating apoptosis process of P53 induced bcl-2.
