脑室内低温对兔脑缺血后神经细胞凋亡及P53蛋白表达的影响 被引量:4

摘要 目的探讨脑室内低温对新西兰白兔局部脑缺血损伤后神经细胞凋亡及P53蛋白表达的影响。方法采用线栓法制作新西兰白兔大脑中动脉闭塞(MCAO)局部脑缺血模型,缺血时间24h。雄性新西兰白兔24只,随机分成常温组和亚低温组,2组再随机分成2,3,5-三苯基氯化四氮唑(TTC)组和凋亡组。观察动物行为、TTC染色检测脑梗死体积、TUNEL染色观察细胞凋亡、免疫组化染色观察P53蛋白的表达水平。结果与常温组比较,亚低温组动物肢体偏瘫程度轻、脑梗死体积小、缺血周围神经细胞凋亡少,P53蛋白表达降低。结论亚低温神经保护机制涉及P53蛋白的表达,脑室内低温是治疗急性脑缺血的一个新方法。 Objective To explore the effects of intro-ventricle hypothermia on neurons apoptosis and P53 gene exprossion in rabbit model of cerebral ischemia. Methods The rabbit model of middle cerebral artery obstruction was made by intraluminal thread approach for 24 hours. 24 New Zealand male rabbits were randomly divided into common temperature and hypothermia groups, each group being randomly divided into 2,3,5-Triphenyhetrazolium chloride and apoptosis subgroups to evaluate the performance of rabbits by Longa score, the infarction volume of the brain by TTC dye ,the apoptosis cells by TUNEL dye and the expression level of P53 gene by immunohistocbemistry. Results The animals in intro-ventricle hypothermia group had higher performance score, smaller infarction volume ,fewer apoptosis cells around the ischemic core and lower P53 gene expression compared with those of normal temperature group. Conclusions The neuroprotective mechanisms of hypothermia include P53 gene expression, intro-ventricle hypothermia approach is a novel method of treating acute cerebral ischemia, which merits further investigation.
