微波辐射对大鼠海马线粒体COX基因表达的影响 被引量:3

The changes of the COX gene expression in mitochondria in hippocampus of rats after exposure to microwave radiation

摘要 目的探讨微波辐射后大鼠海马线粒体呼吸链相关基因细胞色素C氧化酶(COX)表达的变化。方法采用30mW/cm^2微波辐射30只雄性Wistar大鼠,于辐射后6h、1d、3d和7d活杀取海马,采用逆转录-聚合酶链反应(reverse transcription—polymerase chain reaction,RT—PCR)和图像分析检测大鼠海马组织COX亚基Ⅰ、Ⅱ和ⅣmRNA表达变化,通过Western Blot和图像分析检测大鼠海马组织COXⅠ蛋白表达变化。结果辐射后大鼠海马COXⅠ与ⅡmRNA变化规律相似,即与假辐射组相比,于辐射后6h减少,1d增加,3d和7d见恢复;COXⅣmRNA于辐射后1d表达增加,3d和7d见恢复;大鼠海马COXⅠ蛋白于辐射后6h略降低,1d显著降低(P〈0.05),3d见增加,7d基本恢复。结论微波辐射后大鼠海马线粒体呼吸链相关基因COX表达降低,可能参与其能量代谢障碍的病理生理过程。 Objective To study gene expression of cytochrome c oxidase (COX) subunit in hippocampus of rats after microwave exposure. Methods Thirty male Wistar rats were exposed to microwave radiation at 30 mW/cm^2, Rats were sacrificed at 6 h, 1 d, 3 d and 7 d after exposure. Reverse transcription-polymerase chain reaction (RT-PCR) and image analysis were used to study the expression of COX Ⅰ, Ⅱ and Ⅳ mRNA. The expression of COX Ⅰ protein was detected by Western Blot and image analysis. Results The changes of COX Ⅰ and ⅡmRNA had the similar regular pattern which were decreased at 6 h after exposure and increased at 1d, while retrieved 3 d after. The expression of COX Ⅳ mRNA was increased at 1 d, while retrieved 3 d and 7 d after. The expression of COX Ⅰ protein was a little decreased at 6 h, and decreased at I d significantly ( P 〈 0.05 ) , while increased at 3 d and retrieved at 7 d. Conclusion It manifested that COX Ⅰ, II and Ⅳ of the mitochondria influenced energy production and participated in the process of dysbolism.
