摘要
以从荷兰引进的4个郁金香品种病株为材料,利用马铃薯Y病毒的简并引物和黄瓜花叶病毒CP基因的一对特异引物,通过RT-PCR技术分别扩增出与预期大小相一致的1800 bp和840 bp左右的片段,而对照无任何产物。感受态转化后,进行测序,结果表明田野、多道多、雪莉和克思尼4个品种中均能检测到郁金香碎色病毒和黄瓜花叶病毒。
Four tulip cultivars were introduced from Holland. Degenerate primers for potato Y virus and specific primer for cp gene of cucumber mosaic virus was designed , The 1800bp and 840bp DNA fragement was amplified in the four tulip cultivars by the reverse transcription and polymerase chain reaction (RT-PCR), while no fragment was obtained from the control . The sequence result revealed that both tulip breaking virus and cucumber mosaic virus were detected from Tianye, Duodaoduo, Xueli, Kesini respectively.
