摘要
为了探讨微波辅助提取蓝、紫粒小麦麸皮中色素的方法,以两个蓝粒小麦品种和两个紫粒小麦品种的麸皮为原料,以酸性乙醇为浸提剂,利用格兰仕WD900I型家用微波炉,研究了微波功率、提取时间、料液比(原料与提取剂的比例)和提取剂浓度等单因素对提取率的影响,并根据单因素试验结果,设计了四因素三水平正交试验。结果表明,4个因素对酸性乙醇微波辅助提取蓝、紫粒小麦色素的影响的大小顺序均为微波功率>提取时间>料液比>提取剂浓度;每次加热3瓶(每瓶20或24 mL)的最佳提取条件,微波功率均为450 W,加热时间均为90 s(间断加热,将最高加热温度控制在≤75℃),溶剂均为70%乙醇溶液,料液比,蓝粒为1 g∶10 mL,紫粒为1 g∶12 mL。
Studies on extraction of bran pigment of wheat with blue grain or violet grain by microwave is important. The bran of two wheat lines with blue grain and two wheat lines with violet grain were studied by acid-ethnol as extraction solution and microwave as assisted-extraction method with the single factor experiment,including microwave power, extration time, solution cocentration and sample-solution ration. The L9 (3^4) orthogonal experiment was conducted based on the results of the single factor experiment. The results showed that the rank of factors extracting pigment of blue-grain wheat and violet-grain wheat by acidic-athanol assisted with microwave were microwave power, extration time, sample-solution ration and solution cocentration. When three 20-24 mL bottles of samples in extration solution were heated, the optimized extraction condition was that microwave power 450W, time 90s(heated intermittently and the maximum heating temperature ≤75℃) and solution of 70% acid-ethnol, and sample-solution ratio of 1 g : 10 mL for blue-grain wheat and 1 g : 12 mL for violetgrain wheat.
