摘要
目的:观察针刺对家兔实验性动脉再狭窄的防治作用并探讨其作用机理。方法:应用PTCA球囊导管对实验兔行双侧髂动脉内膜剥脱以建立动脉粥样硬化模型,术后1月髂动脉造影示50%以上狭窄者再行PTCA球囊扩张术,以建立再狭窄模型。模型成功后,将造模动物随机分成针刺实验组、普罗布考组、模型组,每组8只。自术前2天至术后14天为1疗程,针刺内关(双)、厥阴俞(双)、心俞(双)、膻中、足三里(双)等穴。髂动脉取材,石蜡切片,TUNFL染色,DAB显色,苏木素复染。观察各组动脉内皮细胞凋亡率及Bax、Bcl-2基因蛋白表达。结果:针刺组内皮细胞凋亡率及Bax基因表达明显高于模型组(P<0.01),针刺后Bax/Bcl-2比值明显升高。结论:针刺能促进兔血管再狭窄模型平滑肌细胞凋亡,调整Bax/Bcl-2比值,从而达到防治PTCA后动脉再狭窄的作用。
Objective:To observe the effects of acupuncture in preventing and treating artery restructure of rabbits. Methods:Artery restructure models were established by PTCA sacculus proprius to denudation arteria inner membrane two sides.Rabbits were divided into four groups:normal,model,acupuncture and probucol groups.It was a treatment course from the first 2 days preoperation to the 14th day post operation.Acupuncturing aoupoints such as neiguan,jueyinshu,xinshu,tanzhong and zusanli,etc.were taken.The rate of apoptosis in arterial endothelial cells and the expression of Bax and Bcl-2 genes protein were determined in each group. Results:The apoptosis rate of endothelial cells and expression of Bax and Bcl-2 genes protein were obviously higher than those of normal group,they were lowered significantly in the acupuncture group,there was obvious difference between the acupuncture group and the model. Conclusion:Acupuncture can restrain the VSMC apoptosis in artery restructure rabbit models,which may be one of the mechanism of its pre- venting PTCA.
