PDTC对TGF-β_1诱导的大鼠纤维化肺成纤维细胞转分化的影响 被引量:2

摘要 目的:通过核因子-κB(NF-κB)抑制剂二硫代氨基甲酸吡咯烷(PDTC)探讨NF-κB对转化生长因子-β1(TGF-β1)诱导的肺成纤维细胞转分化的影响。方法:采用体外细胞培养,免疫细胞化学技术,流式细胞术等技术,检测PDTC对TGF-β1诱导的成纤维细胞表达α-平滑肌肌动蛋白(α-SMA)的影响。结果:TGF-β1诱导肺成纤维细胞转分化为肌成纤维细胞(MF),表达α-SMA增强(P<0.05),PDTC抑制了TGF-β1的诱导作用(P<0.05),单独应用PDTC对肺成纤维细胞转分化无影响。结论:NF-κB促进TGF-β1诱导肺纤维化模型中肺成纤维细胞转分化成MF。 Objective:To investigate the effects of NF-κB on TGF -β1-induced differentiation of pulmonary fibroblasts to myofibroblasts by using PDTC(inhibitor of NF-κB). Methods: Cell culture in vitro, immunocytochemical staining and flow cytometry were used to assess the effect of PDTC on TGF-β1-induced α-SMA expression in Pulmonary fibroblasts. Results; The expressions of α-SMA were increased in TGF β1-group compared with those in sham group. The expressions of α-SMA in PDTC + TGF-β1 group were lower than those in TGF-β1 group, PDTC alone has no effect on the expression of α- SMA. Conclusion: NF-κB promotes the differentiation of pulmonary fibroblasts induced by TGF-β1.
