葡聚六糖诱导植物防卫反应的信号传导 被引量:1

摘要 以烟草悬浮细胞和黄瓜为试材,采用常规试验方法测定防卫反应相关指标,探讨葡聚六糖诱导植物后防卫反应信号传导的途径。结果表明:葡聚六糖诱导后,烟草悬浮细胞在50min时pH值达峰值,为5.75;60min时一氧化氮(NO)含量达峰值,为38.4μmol.mL-1,80min时苯丙氨酸解氨酶(PAL)活性达到峰值,为3.06U·g-1.FW-1;30min时过氧化氢(H2O2)含量达峰值,为3.8μmol·g-1.FW-1,同时过氧化氢酶(CAT)活性也达峰值为129.3U·g-1.FW-1。添加不同抑制剂EGTA、新霉素、氯化锂(LiCl)等测定发现,细胞胞内和胞外Ca2+参与调控葡聚六糖诱导的NO释放及H2O2迸发。葡聚六糖诱导黄瓜产生对霜霉病菌的系统抗性,处理叶及其上位叶和下位叶的防效分别为81.34%、52.12%、6.69%。测定黄瓜叶片PAL、H2O2及其清除酶CAT、SOD活性发现,处理叶活性达到高峰之际,其上位叶和下位叶的活性也先后达峰值,此间抗病信号发生了系统传导。 Using tobacco suspension culture and cucumber as materials, the signal transduction pathway of defensive reaction in plant induced with glucohexaose by means of conventional test method was studied to detect relative targets.The results showed that maximum of pH was 5.75 by 50 min, and content maximum of NO was 38.4μmol·mL^-1 by 60min, and high active of PAL was 3.06 U·g^-1·FW^-1 by 80 min, and content maximum of H2O2 was 3.8μmol·g^-1·FW^-1by 30min, while high active of CAT was 129.3 U·g^-1·W^-1 on tobacco suspension cells treated with glucohexaose. By adding different inhibitors such as EGTA, neomycin, LiCl and so on, it was indicated that intracellar and extracellar Ca^2+ might take part in the role in regulating release of nitric oxide and active oxygen burst on tobacco suspension cells. Systemic disease resistance of cucumber to Pseudoperonospora cubensis were induced by glucohexaose, and protective effect were 81.34%, 52.12% and 6.69% respectively on leaves treated, upper leaves and lower leaves from the treated leaves. When active of PAL, H2O2, CAT, SOD were up to high on leaves treated, active of them were also up to high on upper leaves and lower leaves from the treated leaves in cucumber, here occurred signal trans- duction of systemic disease resistance.
