摘要
目的:了解兔脑缺血后再灌注时神经肽分泌的变化和观察再灌注后实施头部重点低温对这些改变的影响。方法:采用新西兰兔56只,8只为常温对照组(Ⅰ组),其余动物先用“四血管”式造成缺血30分钟,再分成常温再灌注和头部重点低温组。两组均分再灌注30、180和360分钟三个亚组,各亚组动物均为8只。头部低温在再灌注开始后一分钟开始实施,脑组织中神经肽含量采用放免法测定。结果:与非缺血组比较,Ⅱ组三种神经肽含量在所测的再灌流的三个时段均明显下降(P<0.01),而低温组加压素(AVP)和β-内啡肽(β-EP)含量无明显下降(P>0.05),血管活性肠肽(VIP)含量在再灌注30和180分钟有明显下降(P<0.01);与常温再灌注组相比,头部低温治疗后三种神经肽的含量均有明显回升(P<0.01)。结论:再灌流时兔脑神经肽分泌紊乱,再灌流后实施低温仍能有效地减轻这种紊乱程度。
Objective:To investigate the changes of endocrine of neuropeptides in hypotholamus during reperfusion,and observe the effects of selective head cooling on these changes in rabbits Method:Fifty six New Zealand rabbits were randomly allocated into three groups,group Ⅰ:normal control (n=8),in other groups complete cerebral ischemia(CCI)was induced with the four vessel model,and reperfusion lasted 30,180 and 360 minutes in three subgroups of group Ⅱ or group Ⅲ respectively Selective head cooling in group Ⅲ was iniated at the begining of reperfusion (28℃,surface cooling method) The levels of neuropeptides(β endphine,vasopress,vasoactive intestinal peptide)were determined by radioimmunoassay Result:Compared with the controls,the levels of three neuropeptides were significantly decreased at three reperfusion periods in group Ⅱ(P<0 01);levels of vasopress and β endphine unchanged in group Ⅲ(P>0 05),while levels of vasoative intestinal peptide were significantly decreased at 30 and 180 minutes of reperfusion(P<0 01) Compared with group Ⅱ(normothermic reperfusion),the levels of three neuropeptides significantly rose(P<0 01) in group Ⅲ following selective head cooling Conclusions:The endocrine of neuropeptides in hypothalmus becomes disturbed during reperfusion,and selective head cooling iniated at the begining of reperfusion has markedly ameliorated effects on these disorders
