摘要
为探讨乳腺癌组织中p53基因丢失与p53mRNA表达的相关性,应用Southern杂交及反转录-定量PCR技术对47例乳腺癌组织进行了检测。结果发现:p53基因杂合性缺失率为34.0%(16/24);在乳腺正常腺体有中度p53mRNA表达,而在肿瘤组织中有40.4%(19/47)患者伴p53mRNA高表达。p53基因缺失与p53mRNA高表达之间呈显著性相关(P<0.01)。提示:p53基因在人类乳腺癌形成和肿瘤演进过程中起重要作用。反转录一定量PCR具有快速、简单、敏感、无放射性污染等优点,适于临床应用。
p53 gene deletion and p53 mRNA expression were examined in 47 cases of breast cancer by using Southern blotting and RT/PCR techniques. p53 gene was found to be partially deleted in 34.0% (16/47) of breast cancer tissues. In the normal breast gland, there was moderately p53 mRNA expression. In the tissues of carcinomas, 40.4% (19/47) of the cases were companied with p53 mRNA overexpression. A highly significant correlation between p53 gene deletion and p53 mRNA expression was found ( P<0.01) . The data indicate that p53 gene may play an important role in the occurrence and progression of breast cancer. The RT/PCR technique is a quick, simple and sensitive method and suitable for widely clinical use.
