摘要
目的:探讨光动力作用(PDT)对人乳腺癌细胞Bax和Bcl-2的影响。方法:体外培养的人乳腺癌细胞MDA-MB-231,分别在PDT作用不同时间后,用免疫组织化学法检测促凋亡蛋白Bax和抗凋亡蛋白Bcl-2蛋白表达情况。结果:Bax蛋白表达在所有组无明显差异(P>0.05),Bcl-2蛋白表达自PDT后6h起显著下降(P<0.01);PDT后6hBax/Bcl-2比值依次递增,且有统计学意义(P<0.05)。结论:光动力作用可诱导人乳腺癌细胞发生凋亡,其机制可能与Bax和Bcl-2蛋白表达的比值有关。
Objective :To explore the effects of photodynamic therapy(PDT) on Bax and Bcl -2 proteins in human breast cancer MDA- MB -231 cells. Methods. Bax and Bcl -2 proteins were detected for PDT by immunocytochemistry. Results: The expressed Bax oncoprotein percent was no difference among all group (P 〉 0.05 ). The percent Bcl -2 protein was significantly decreased since PDT acted 6h later (P 〈 0.01 ). After PDT acted 6h, the Bax/ Bcl - 2 ratio of experimental group was significantly raised with the time prolongation of HpD - PDT ( P 〈 0.05 ). Condusion: Apoptosis could be induced by PDT in vitro, and may be associated with the ratio of Bax/Bcl -2 .
