大鼠Otos基因对碱性成纤维细胞生长因子表达的调控

摘要 目的:探讨Otos基因的表达对碱性成纤维细胞生长因子(bFGF)的调控作用,以揭示Otos基因和bFGF在耳聋病理生理过程中的联系和作用。方法:用含有大鼠Otos基因的腺病毒载体Ad-Otos转染培养的大鼠耳蜗螺旋韧带成纤维细胞,应用RT-PCR技术从转染细胞中检测Otos基因和bFGF基因的表达,用Westernblot方法检测转染细胞中的bFGF蛋白的表达。结果:经过转染Ad-Otos后,成纤维细胞中的bFGF显著上调。结论:Otos基因的表达可能对bFGF表达有影响,并有可能对内耳有保护作用。 Objective: To investigate if there might be any influence of Otos gene expression on the regulation of basic fibroblast growth factor ( bFGF), and to reveal the possible relationship and interactions between them and the path-physiological course of epicophosiss. Methods.. Cultured spiral ligament fibrocytes of SD rats were transfected with different expression vectors including Ad-Otos which contains Otos gene, AD-GFP containing green flourescence protein gene, and DMEM respectively, and, then, mRNA of Otos and bFGF as well as protein of bFGF were assessed with RT-PCR and Western blot. Results: Compared with those in the control ceils, mRNA expression of Otos and mRNA and expressed protein of bFGF in the fibrocytes transfected with Ad-Otos evidently increased. Conclusions: Elevated expression of Otos gene might have an influence on expression of bFGF and have potential protective effects to inner ear.
