人胰腺癌不同转移潜能细胞株的分离筛选和鉴定 被引量:2

摘要 目的获得不同转移潜能的胰腺癌细胞株。方法通过有限稀释法获得人胰腺癌细胞系AsPC-1中不同的单细胞克隆细胞瘤,经过侵袭实验初筛后选择转移潜能不同的2株亚细胞株N1的N7,利用细胞生长曲线测定、软琼脂克隆形成实验、细胞周期检测、裸鼠种植癌转移状态来分析它们的生物学性质。结果N7体外生长速度明显快于N1;N7软琼脂克隆形成率为109±13,明显高于N1的26±5(P〈0.05);N7的G2/M比率为26.3±1.9,明显高于N1的8.3±1.3(P〈0.01);N7裸鼠体内种植癌肝转移率为100%,明显高于N1的0。结论从同一亲本筛选出转移潜能高低的2个亚细胞系,N7为高转移潜能亚细胞系,N1为低转移潜能亚细胞系。 Objective To retrieve human pancreatic carcinoma cell subclones with different potential of metastasis.Methods Different monocell clones were retrieved by the limited dilution method from the human pancreatic carcinoma cell line AsPC-1.After preliminary screening by in vitro invasion assay,we selected one cell lines as high metastasis potential cell subclones(N7) and one cell subclones as low metastasis potential cell subclones(N1).We analyzed their biological features by tests of cell growth curve assay,soft agar cloning technique,flow cytometry and the status of metastasis in nude mice.Results The two subclones with the different metastasis potential were retrieved:low potential subclone N1 and high potential subclone N7.N7 grew faster than N1,and the cloning efficiency rate was 26 ± 5 for N1 and 109 ± 13 for N7.The rate of G2/M was 8.3 ± 1.3 and 26.3 ± 1.9(P 〈 0.01) for N1 and N7,respectively.The artificial liver metastasis rate of N1 and N7 in nude mice was 0% and 100%,respectively.Conclusions These two subclones derived from the same mother cell line and their difference mainly lies in the different metastatic phenotype.The study will facilitate the understanding of the metastatic mechanism of pancreatic carcinoma.
