摘要
用全胚胎培养并结合病理检查探讨了不同浓度砷对离体胚胎组织器官的影响。镜下结果表明,砷对脑部、神经管、心脏、卵黄囊、肢芽、体节等组织器官的形态分化均有明显抑制作用,呈现剂量反应关系。0.5μg/ml砷仅对脑部有轻度损害,可见内皮层和边缘层细胞脱落,说明脑是砷毒作用的一个敏感靶器官。砷≥1.0μg/ml时,脑和神经管的室管膜层细胞数量减少、排列紊乱、变性坏死;同时可见卵黄囊三层膜结构紊乱、微血管数量减少;心脏。
In the present study,the method of whole embryo culture (WEC) was used to expose 9.5 day rat embryos to arsenic trioxide (0~2.0μg/ml) in serum from same Sprague Dawley rat.Embryos were examined after 48h in WEC in vitro for malformations,et al.Then,embryos were dissected,and serial optical sections (8u) were obtained and observed by light microscopy.The results revealed arsenic inhibited morphological differentiation of different organs,including brain,neural tube,heart,visceral yolk sac (VYS),limb bud,somite,et al.in a dose dependent manner.The cell exfoliation of epithelial and edge layer was only discovered in 0.5μg/ml group,which demonstrated that brain was a susceptible target tissue.At≥1.0μg/ml groups not only epicytes of brain and neural tube exfoliated but ependymocytes were out of order,and degenaration and necrosis of some ependymocytes were found.At the same time,those pathological changes of VYS,heart,limb bud were observed,for example,the structural disorder of VYS and so on.
